Mechanisms of Mediator-Dependent Transcription Activation

The Mediator complex is an essential regulator of gene expression that relays signals from gene-specific transcription factors (TFs) to the core transcription machinery. As a large, dynamic, and modular protein assembly, Mediator consists of more than twenty subunits that harbor numerous TF-binding interfaces, and Mediator can undergo extensive conformational rearrangements in response to TF binding. My project aims to characterize the molecular interactions and mechanisms of TF- and Mediator-dependent transcription activation using in vitro biochemical assays, real-time microscopy, and cell-based methods, with the goal of advancing our understanding of transcription regulation and enabling its precise modulation.
